Defensin beta 136 (DEFB136)

Target
DEFB136
Molecular classification
Antimicrobial peptide, Beta defensin family, Cysteine-rich cationic protein, Secreted peptide
01

Overview

Defensin beta 136 (DEFB136) is a secreted, cysteine-rich cationic antimicrobial peptide belonging to the beta defensin family, located in the defensin gene cluster on human chromosome 8p23. Beta-defensins, including DEFB136, act as components of the innate immune system, exerting their effect primarily through broad-spectrum antimicrobial activity against bacteria and fungi, and may also modulate immune cell behavior. DEFB136 is reported to be preferentially expressed in the male reproductive tract, where it contributes to both local immunity and immunomodulation. No drugs are currently known to act specifically via DEFB136, but its properties are of research interest for anti-infective and immunomodulatory strategies.

Mechanism of action

Direct disruption of microbial membranes via cationic interaction and pore formation; Immune signaling/chemotactic activity for recruiting immune cells, as described for other beta-defensins

03

Biological functions

Antimicrobial activity (active against Gram-negative and Gram-positive bacteria)Antifungal innate immune responseImmunomodulatory role (may influence immune cell recruitment and modulate inflammation)Lipopolysaccharide binding
04

Disease associations

Infection (protection against microbial colonization, especially in mucosal surfaces)Inflammation (potential roles in mucosal inflammatory states, but specific associations for DEFB136 are limited)Other (potentially male reproductive immunity and fertility, based on expression patterns)
05

Safety considerations

No specific concerns identified for DEFB136 pharmacological modulationGeneral challenges for defensin-based therapy include stability, selectivity, and potential cytotoxicity at elevated doses
