Degraded vascular elastin refers to the fragmented state of the elastin protein within the extracellular matrix of blood vessels, typically resulting from chronic inflammation and the action of matrix metalloproteinases (Sinha et al., 2014). In healthy tissue, elastin provides essential elastic recoil and structural integrity; however, its degradation is a hallmark of vascular pathologies such as abdominal aortic aneurysms and atherosclerosis (Isenburg et al., 2007). This damaged matrix serves as a unique therapeutic target because it exposes specific hydrophobic domains and epitopes not accessible in intact elastin (Nosoudi et al., 2016). Current pharmacological strategies involve using polyphenols like pentagalloyl glucose (PGG) to cross-link and stabilize these fragments, thereby halting disease progression (Vyavahare et al., 2014). Additionally, degraded elastin is utilized as a docking site for targeted nanoparticle delivery systems designed to release drugs specifically at the site of vascular injury (Shokeen et al., 2011). Monitoring the extent of degradation often involves measuring biomarkers like desmosine or elastin-derived peptides in the blood (Ma et al., 2007).
Stabilization of the extracellular matrix through cross-linking of elastin fibers and inhibition of elastolytic enzymes, or serving as a localized anchor for targeted drug delivery systems (Isenburg et al., 2007; Nosoudi et al., 2016).
