Overview

Deleted in lymphocytic leukemia 7 (DLEU7) is a protein-coding gene located at chromosome 13q14, a region frequently deleted in B-cell chronic lymphocytic leukemia (CLL)[1][2][5][6]. DLEU7 encodes a protein of 221 amino acids, highly conserved in mammals, with a proposed role as a tumor suppressor[6]. Its biological function centers on the inhibition of NF-κB and NFAT signaling pathways through direct physical interaction with TACI and BCMA, two members of the TNF receptor superfamily involved in the survival and proliferation of B-lymphocytes[2][4]. Loss or deletion of DLEU7 leads to enhanced signaling via these pathways, promoting cell proliferation and survival, and is implicated in the pathogenesis of CLL and other hematologic malignancies[2][4][6]. DLEU7 expression has primarily cytoplasmic localization, and its repression (often through promoter methylation) correlates with increased risk and progression of chronic lymphocytic leukemia[6]. The gene is primarily associated with tumor suppressive function and is not a direct therapeutic target or receptor; consequently, no known drugs target DLEU7 specifically nor is there evidence for established mechanisms of action or notable safety concerns relevant to drug modulation[2][4][6].
