DLK1 encodes a transmembrane protein that regulates cell growth and differentiation, particularly adipogenesis. It exists in both membrane-bound and soluble forms, the latter generated by proteolytic cleavage. While it belongs to the Notch/Delta/Serrate family, it lacks the canonical DSL domain, suggesting non-canonical signaling mechanisms. DLK1 is implicated in cancer, where its overexpression is associated with poor prognosis and may mark cancer stem cells.
ADAM17/TACE inhibition may indirectly affect DLK1 signaling, as it prevents the shedding of soluble DLK1.
