DTX3L is an E3 ubiquitin ligase that plays crucial roles in DNA damage repair, antiviral responses, and signal transduction. It uniquely can ubiquitinate both protein substrates (including histones) and single-stranded nucleic acids. The protein forms important complexes with PARP9 and is over-expressed in chemotherapy-resistant lymphomas, making it a potential therapeutic target for cancer treatment.
