Overview

Dematin actin-binding protein (DMTN) is a cytoskeleton-associated protein that binds and bundles F-actin, playing a pivotal structural role especially in erythrocytes by stabilizing the spectrin-actin cytoskeleton and anchoring it to the plasma membrane in a phosphorylation-dependent manner[1][3]. DMTN exists as a trimer, generated by alternative splicing, and contains a core domain and a C-terminal "headpiece" domain similar to villin, with the headpiece directly mediating actin binding and bundling[2][4]. Its actin-bundling activity is dynamically regulated by phosphorylation (notably by cAMP-dependent protein kinase A); when phosphorylated, its ability to promote bundle formation is diminished[1][3][4]. DMTN is required for maintaining erythrocyte shape, membrane elasticity, and mechanical properties; loss or mutation leads to disorders like hereditary spherocytosis and compensated anemia. Beyond erythrocytes, DMTN functions in other cell types, modulating cytoskeletal dynamics, membrane stability, and cell signaling, including roles in platelets (calcium mobilization and secretion) and suppression of cancer cell invasion by controlling actin rearrangement machinery. Loss or epigenetic silencing of DMTN has been implicated in cancer progression, especially via pathways involving Rac1-dependent cytoskeletal remodeling[1][3][4].
