Dendritic cell (DC) activation is the process by which immature DCs transform into mature antigen-presenting cells, initiating adaptive immune responses. This involves recognition of pathogens or danger signals via PRRs, antigen internalization, upregulation of surface molecules (MHC II, CD80/CD86/CD40, CCR7), and secretion of cytokines like IL-12. The process is essential for bridging innate and adaptive immunity and can be manipulated for vaccine design, immunotherapy, and tolerance induction.
Dendritic cell activation is triggered by the recognition of PAMPs/DAMPs by PRRs, leading to upregulation of MHC molecules, costimulatory molecules, and chemokine receptors. This enables antigen presentation, T cell activation, and migration to lymphoid tissues, shaping the adaptive immune response. Drugs can modulate this process by directly stimulating DCs (TLR agonists), indirectly activating them through cytokine release, or suppressing their function (immunosuppressants).
