QS-21 is a natural saponin derived from *Quillaja saponaria* used as a potent vaccine adjuvant. Upon administration—often in liposomal formulations—QS-21 is rapidly internalized by antigen-presenting cells, especially dendritic cells and lymph node-resident macrophages. In the context of vaccines, QS-21 promotes inflammasome activation (notably via NLRP3), which leads to caspase-1-dependent release of IL-1β and HMGB1, driving local recruitment and activation of dendritic cells. The enhanced function of dendritic cells supports robust antigen presentation, driving both humoral and cellular adaptive immune responses. QS-21’s clinical utility is significant for vaccines against cancer, infectious, and some neurodegenerative diseases, though toxicity and chemical instability present challenges for formulation and dosing[1][2][3][4][5]. Note: “Dendritic cell activation via QS-21” is not a receptor, enzyme, transporter, or canonical molecular target, but a process describing how QS-21 exerts its adjuvant effect through immune cell activation.
QS-21 enhances antigen uptake and presentation by dendritic cells via endocytosis and activation of the NLRP3 inflammasome, resulting in secretion of IL-1β and IL-18[1][3][5]. Activation leads to mixed Th1/Th2 responses: both cytotoxic T cell activation and antibody response[2][5]. Induction requires MyD88-dependent signaling; Caspase-1 activation is critical for IL-1β secretion and immune cell recruitment in draining lymph nodes[1][3]. QS-21 is rapidly captured by lymph node macrophages, which then orchestrate dendritic cell recruitment and activation[1].
