The dendritic cell antigen presentation machinery (DC APM) is a complex multi-component system responsible for the capture, processing, and display of antigens to T-cells. This machinery includes various proteins such as Major Histocompatibility Complex (MHC) class I and II molecules, the Transporter associated with Antigen Processing (TAP), tapasin, and various proteasomes and cathepsins. By converting intracellular or extracellular proteins into peptide fragments and loading them onto MHC molecules, dendritic cells serve as the primary bridge between innate and adaptive immunity. In many cancers, the APM is frequently downregulated or mutated, allowing tumor cells to evade immune detection. Therapeutic strategies often aim to restore or enhance this machinery using toll-like receptor (TLR) agonists, cytokines like Interferon-gamma, or dendritic cell-based vaccines to stimulate a robust cytotoxic T-lymphocyte response.
Modulation of the antigen presentation machinery typically involves the use of adjuvants or cytokines to upregulate the expression of MHC molecules, transporters (TAP), and costimulatory signals, thereby enhancing the visibility of pathogens or tumor cells to the adaptive immune system.
