Dendritic cell antigen-presenting ability refers to the functional capacity of dendritic cells (DCs) to capture, process, and display antigens to T cells, thereby initiating an adaptive immune response (PMC2691475). As 'professional' antigen-presenting cells, DCs bridge innate and adaptive immunity by displaying antigen-derived peptides on Major Histocompatibility Complex (MHC) molecules and providing essential co-stimulatory signals via surface proteins such as CD80 and CD86 (NIH/PMC). In various disease states, particularly within the tumor microenvironment, this ability is frequently suppressed by factors like vascular endothelial growth factor (VEGF), interleukin-10 (IL-10), and transforming growth factor-beta (TGF-β), leading to immune evasion and tolerance (Taylor & Francis). Therapeutic strategies, including VEGF inhibitors like bevacizumab or the administration of maturation cytokines, aim to restore or enhance this ability to promote anti-tumor immunity. Consequently, monitoring this functional phenotype through markers like HLA-DR and CD83 is critical for assessing the efficacy of immunotherapies and understanding immune dysfunction in chronic infections and trauma (PubMed).
Pharmacological agents modulate this functional capability by inhibiting immunosuppressive factors in the microenvironment (e.g., VEGF) or by providing exogenous maturation stimuli (e.g., cytokines and TLR agonists) to enhance MHC expression and co-stimulatory molecule density on the dendritic cell surface.
