Overview

Dendritic cell (DC) co-stimulatory receptors are a heterogeneous group of cell-surface proteins that provide the secondary signals necessary for the activation, survival, and differentiation of T-cells during antigen presentation, often referred to as Signal 2. Key members include the B7 family proteins, such as CD80 and CD86, which bind to CD28 or CTLA-4 on T-cells, and members of the tumor necrosis factor receptor (TNFR) superfamily, such as CD40, OX40L (CD252), and 4-1BBL (CD137L) (Source: UniProt; Janeway's Immunobiology). These receptors play a pivotal role in the immunological synapse, where their engagement determines whether a T-cell becomes effector-active or enters a state of anergy or tolerance (Source: Nature Reviews Immunology).
