Dendritic cell (DC) differentiation refers to the process by which precursor cells acquire the specialized features of dendritic cells, a critical component of the immune system responsible for antigen presentation and T-cell activation. DCs originate from hematopoietic stem cells (HSCs) in the bone marrow and undergo a multistep differentiation cascade involving several intermediate progenitor stages. The process is regulated by cytokines (GM-CSF, TNF-alpha), transcription factors (BATF3, IRF4, E2-2), and metabolic pathways (mTOR). Immature DCs mature upon encountering antigens, upregulating MHC and co-stimulatory molecules, and migrate to lymph nodes to activate T-cells.
