The dendritic cell (DC) maturation state refers to the functional and phenotypic status of a dendritic cell as it transitions from an immature, antigen-capturing cell to a mature, antigen-presenting cell capable of priming T cells. This process is central to the DC's role as a bridge between innate and adaptive immunity. The maturation state determines whether DCs induce immune activation or tolerance; thus, it is critical in shaping immune responses such as Th1 vs. Th2 polarization or tolerance induction. Key features of maturation include increased expression of MHC class I/II and costimulatory molecules (CD80/CD86/CD40), upregulation of CCR7, and secretion of inflammatory cytokines.
