Dendritic cell (DC) maturation induced by the heat shock protein gp96 (grp94) complexed with peptides is a critical process for initiating adaptive immune responses. The gp96-peptide complex acts as an adjuvant and delivers antigenic material to DCs, triggering their maturation. This maturation involves the upregulation of MHC class II and costimulatory molecules (CD80, CD86, CD40, CD83), secretion of pro-inflammatory cytokines (IL-12, TNF-α), and enhanced ability to stimulate naïve T cells. Gp96-peptide complexes are being explored as cancer vaccine candidates due to their ability to stimulate cytotoxic T lymphocytes against tumor-associated antigens.
Gp96-peptide complexes bind to receptors on dendritic cells, leading to receptor-mediated uptake and subsequent activation of intracellular signaling pathways. This results in the upregulation of MHC molecules, costimulatory molecules, and pro-inflammatory cytokine secretion, ultimately leading to DC maturation and T cell priming.
