Overview

Dendritic cell-mediated antigen presentation pathways are the primary mechanisms by which the immune system identifies and responds to foreign or abnormal antigens (StatPearls, 2023). Dendritic cells (DCs) act as professional antigen-presenting cells that capture antigens, process them into peptides, and present them on Major Histocompatibility Complex (MHC) molecules to T-cells (Roche & Furuta, 2015). This process is critical for the activation of CD4+ T helper cells via MHC class II and CD8+ cytotoxic T cells via MHC class I, the latter often occurring through a specialized process known as cross-presentation (Embgenbroich & Burgdorf, 2018). These pathways are central to the development of adaptive immunity against pathogens and tumors (Wculek et al., 2020). Therapeutically, these pathways are targeted using DC-based vaccines, such as Sipuleucel-T, or adjuvants like TLR agonists to enhance immune recognition of cancer cells. Conversely, inhibiting these pathways is a strategy for managing autoimmune diseases and preventing organ transplant rejection.
