Overview

Dendritic cell pattern-recognition and antigen-presentation pathways represent the integrated biological processes by which dendritic cells (DCs) detect pathogens and initiate adaptive immune responses. DCs utilize various pattern recognition receptors (PRRs), such as Toll-like receptors (TLRs) and C-type lectin receptors, to sense pathogen-associated molecular patterns (PAMPs) or damage-associated molecular patterns (DAMPs) (Janeway & Medzhitov, 2002, Annu Rev Immunol). Upon activation, DCs undergo maturation, characterized by the upregulation of co-stimulatory molecules like CD80 and CD86 and the processing of captured antigens into peptides (Banchereau & Steinman, 1998, Nature). These peptides are then loaded onto Major Histocompatibility Complex (MHC) class I and II molecules for presentation to CD8+ and CD4+ T cells, respectively (Roche & Furuta, 2015, Nat Rev Immunol). This pathway is a critical bridge between innate and adaptive immunity and is frequently targeted in immunotherapy and vaccine development to enhance anti-tumor or anti-viral responses (Mellman & Steinman, 2001, Cell). Drugs such as imiquimod and various DC-based vaccines (e.g., Sipuleucel-T) aim to modulate these pathways to stimulate robust T-cell mediated immunity (Adams, 2009, Immunotherapy).
