Overview

Dendritic cell receptor DEC-205 (CD205, also known as LY75) is a type I transmembrane endocytic receptor mainly expressed by dendritic cells and is a member of the macrophage mannose receptor family[2][3]. It contains multiple C-type lectin-like domains and mediates the uptake and internalization of a broad range of antigens—both self and foreign—into MHC II-positive endosomal/lysosomal compartments, facilitating antigen processing and cross-presentation to T lymphocytes[4][8]. DEC-205 serves as a key pathway for antigen capture, especially in immune surveillance and tolerance, and specifically recognizes apoptotic and necrotic cells by a pH-dependent mechanism[1]. It also directly binds CpG oligonucleotides (bacterial/viral DNA motifs) and mediates their internalization, which is utilized for activation of dendritic cells and the enhancement of immune responses in vaccine strategies[5][6]. DEC-205 is exploited as a target for the delivery of antigens or adjuvants to dendritic cells to boost immune responses against infections and tumors. Its functions in immune modulation, cross-presentation, and tolerance position it as a promising but complex therapeutic and vaccine target[1][4][5][6][8].
