Dendritic Cell-Specific Intercellular Adhesion Molecule-3-Grabbing Non-Integrin (DC-SIGN/CD209) is a C-type lectin receptor primarily expressed on immature dendritic cells and certain macrophage populations. It functions as a pathogen recognition receptor, binding high-mannose carbohydrates on pathogens like HIV and dengue virus, facilitating their entry into cells. It also acts as a cell adhesion molecule, interacting with ICAM-3 on T lymphocytes to initiate T-cell activation. DC-SIGN ligation can lead to increased anti-inflammatory cytokine release, influencing the tumor microenvironment and potentially promoting tumor progression. Due to its role in both pathogen recognition and immune modulation, it is a target for research in infectious diseases, cancer immunology, and vaccine development.
