Target intelligence / Profile preview
Dendritic cell-specific receptor DEC-205 (DEC-205, also known as CD205 or Ly75) is a type I transmembrane C-type lectin receptor prominently expressed on dendritic cells and cortical thymic epithelial cells[1][4][5][7][9]. Structurally, it features a cysteine-rich N-terminal domain, a fibronectin type II domain, and 10 C-type lectin-like domains, forming two intercalated rings that mediate ligand binding and pH-dependent oligomerization[4][5]. DEC-205 functions as an endocytic receptor specialized for efficient uptake of antigens, processing them in late endosomal/lysosomal compartments, and presenting these antigens on major histocompatibility complex (MHC) class I and II molecules to T cells[1][3][9]. This enables dendritic cells to stimulate immune responses or induce immune tolerance, depending on the context. DEC-205 recognizes cellular material from apoptotic and necrotic cells and is crucial for intrathymic and peripheral tolerance[9]. It has garnered interest as a target for vaccine development and cancer immunotherapy, where antigens or adjuvants are conjugated to anti-DEC-205 antibodies to direct their uptake by dendritic cells, enhancing antigen presentation and immune activation[1][9]. No approved small-molecule drugs target DEC-205 directly, but experimental studies use monoclonal antibodies and CpG oligonucleotides for immunomodulation[4][5][6][9].
Targeted delivery of antigens for enhanced dendritic cell-mediated antigen presentation - Induction of immune responses or immune tolerance, depending on co-administered adjuvants and context[1][3][9]. - Uptake and presentation of apoptotic/necrotic cell-derived antigens for tolerance or cross-presentation[9]. - Antigen internalization via receptor-mediated endocytosis followed by trafficking to antigen-processing compartments[1][3].
