Dendritic cell-specific transcript (DC-SCRIPT), also known as ZNF366, is a transcription factor that is highly and specifically expressed in dendritic cells (DCs). It was originally identified in the literature as a novel dendritic cell-expressed mRNA (DC-mRNA) and plays a critical role in the maturation and functional specialization of DCs, which are essential for initiating adaptive immune responses. DC-SCRIPT acts as a transcriptional corepressor for several nuclear receptors, including the estrogen receptor alpha (ERα), the glucocorticoid receptor (GR), and the progesterone receptor (PR), thereby modulating various signaling pathways. In clinical research, DC-SCRIPT has emerged as a significant prognostic biomarker in breast cancer, where its high expression in tumor cells is associated with improved patient survival and a more favorable clinical outcome. Although it is considered a potential target for immunomodulatory therapies due to its role in DC biology, there are currently no approved drugs that directly target this molecule.
