A dendritic cell stimulant refers to any agent (molecule, ligand, or signal) that activates dendritic cells (DCs), which are key antigen-presenting cells in the immune system. These stimulants trigger DC maturation and functional activation, enabling them to process antigens and present them to T cells, thus bridging innate and adaptive immunity. Examples include pathogen-associated molecular patterns (PAMPs) like LPS and CpG DNA, damage-associated molecular patterns (DAMPs), and cytokines like GM-CSF. Upon stimulation, DCs upregulate surface markers for antigen presentation and co-stimulation, secrete cytokines, and undergo metabolic reprogramming to support their activated state. Manipulating dendritic cell stimulants is central to vaccine adjuvant design and cancer immunotherapy. Agents that stimulate DC maturation enhance immune responses against pathogens/tumors; conversely, inducing tolerogenic states may help treat autoimmune diseases/allergies.
