Upon binding, these receptors trigger intracellular signaling cascades involving molecules like Syk, Raf-1, or MyD88, which ultimately lead to DC maturation and the secretion of cytokines that drive Th2 cell differentiation. This machinery is a critical checkpoint in the sensitization phase of allergic diseases, as it determines whether an inhaled or ingested protein is perceived as harmless or as a threat requiring an IgE-mediated response. While not a single therapeutic target, the components of this machinery are being investigated for their potential to be modulated in allergen-specific immunotherapy or to prevent the onset of allergic sensitization.
