Overview

Dendritic cell (DC) uptake and maturation machinery refers to the integrated system of receptors, signaling pathways, and cellular processes that allow DCs to capture antigens and transition from an immature, phagocytic state to a mature, antigen-presenting state (Banchereau & Steinman, 1998, Nature). This machinery includes C-type lectin receptors (e.g., DEC-205, DC-SIGN) for antigen internalisation and pattern recognition receptors (e.g., Toll-like receptors) that sense danger signals (Mellman & Steinman, 2001, Cell). Upon activation, DCs undergo phenotypic changes, including the upregulation of MHC molecules and costimulatory proteins like CD80 and CD86, which are essential for T-cell activation (Wylie et al., 2019, NIH). In oncology, this machinery is targeted by adjuvants and vaccines to enhance anti-tumor immunity, while in autoimmune contexts, it may be modulated to induce tolerance. Because this term encompasses a wide array of distinct proteins rather than a single molecular entity, it is generally considered a functional pathway rather than a specific therapeutic target.
