The **Dengue virus envelope protein serotype 1** is a major structural glycoprotein of the dengue virus (DENV-1), a member of the flavivirus family that also includes Zika, West Nile, and Japanese encephalitis viruses. The E protein forms homodimers on the surface of the virion and is organized into three domains (DI, DII, DIII), each contributing to the processes of viral attachment, membrane fusion, and entry into host cells by interacting with cellular receptors such as DC-SIGN. Domain III (DIII) serves as the principal receptor recognition and binding motif and is highly immunogenic, making it an important target for vaccine and therapeutic antibody development. The protein’s function is tightly regulated by pH, triggering conformational changes necessary for membrane fusion in the endosome. The envelope E protein is the primary target for neutralizing antibodies, but sequence variation and cross-reactivity can complicate diagnosis and immune protection. No vaccines or approved therapeutics specifically target E protein, but it remains a key focus for antiviral drug and vaccine development
