Dengue virus nonstructural proteins (NS1, NS2A, NS2B, NS3, NS4A, NS4B, and NS5) are a group of seven proteins encoded by the viral genome but not incorporated into the mature virion. These proteins play essential roles in viral replication, assembly of new virions, modulation of host cell processes, and evasion of the immune response. They are considered prime targets for antiviral drug development because they perform essential functions not found in human hosts. Inhibitors targeting these proteins can block multiple stages of the viral life cycle, from polyprotein processing (NS3/NS2B) and genome synthesis/capping (NS5) to assembly/release steps involving other components.
