The dengue virus RNA-dependent RNA polymerase (RdRp), encoded by the NS5 protein, is a crucial enzyme responsible for replicating the viral RNA genome. It catalyzes the synthesis of minus-strand RNA from the positive-strand dengue virus genome, which then serves as a template for producing additional plus-strand genomes. This enzyme is essential for viral replication and transcription, making it a key target for antiviral drug development. Inhibitors can bind allosteric sites within conserved regions to block initiation or elongation steps of replication. The enzyme also participates in capping nascent viral RNAs.
