Overview

The Dengue virus type 1 (DENV-1) envelope (E) glycoprotein is the principal surface protein of the virion and a critical target for the host's immune response. It is a class II viral fusion protein organized into three distinct structural domains: DI, DII (containing the highly conserved fusion loop), and DIII (the primary receptor-binding domain) (UniProt P17763; PubMed 15152264). The protein mediates the initial attachment of the virus to host receptors such as DC-SIGN or heparan sulfate and facilitates pH-dependent membrane fusion within the endosome during viral entry (PubMed 12692220). In the context of therapeutics and vaccine development, the E protein is the primary antigen used to induce neutralizing antibodies. Live-attenuated vaccines like TAK-003 and recombinant subunit vaccines like V180 specifically target the E protein to provide serotype-specific protection (Takeda; PubMed 27156540). However, a major challenge in targeting this molecule is antibody-dependent enhancement (ADE), where sub-neutralizing or cross-reactive antibodies against the E protein can facilitate viral entry into Fc-receptor-bearing cells, potentially exacerbating disease severity (Nature Reviews Microbiology 2020). Consequently, therapeutic strategies focus on engineering antibodies or small molecules that bind with high affinity to the E protein to ensure complete neutralization across different viral strains.
