DENN domain containing 4A (DENND4A) is a protein expressed in humans with a DENN domain structure and functions as a guanine nucleotide exchange factor (GEF) for Rab10, a small GTPase involved in vesicle trafficking. DENND4A was originally identified as a protein binding to the interferon-stimulated response element in the MYC promoter and sometimes called C-myc promoter-binding protein. Its main characterized molecular role is in vesicle-mediated transport, activating Rab10 by promoting the exchange of GDP for GTP. Additionally, evidence suggests DENND4A may influence oncogenic processes by regulating MYC expression. Although linked to certain diseases such as cancer and inherited cardiomyopathy, its direct contribution to disease pathology remains under investigation. No drugs are currently approved to target DENND4A directly, and its role as a biomarker or its safety profile as a therapeutic target have not been established.
