DENN domain-containing protein 1B (DENND1B) is a guanine nucleotide exchange factor (GEF) that activates the small GTPase RAB35, acting primarily in clathrin-mediated endocytosis and endocytic recycling[1][5][7]. It regulates internalization of the T-cell receptor in T helper 2 (TH2) lymphocytes, thereby modulating cytokine production and immune responses[5]. Genetic association studies implicate DENND1B variants in susceptibility to childhood asthma and obesity in both humans and dogs, potentially through effects on energy balance and melanocortin 4 receptor trafficking[1][5][8]. The protein acts as an adapter linking RAB35 activation to clathrin-coated endocytic vesicles and the AP2 adapter complex, playing a key role in vesicular trafficking in immune and other cells[1][5][7]. While DENND1B is not the direct target of approved drugs, its pathway modulation could have relevance for disease risk and therapy in immune and metabolic disorders.
