DENN domain-containing protein 1C (DENND1C, also known as connecdenn 3) is a guanine nucleotide exchange factor (GEF) that primarily functions to activate the small GTPase RAB35 by promoting the exchange of GDP for GTP, regulating endosomal membrane trafficking and actin polymerization[1][4][3]. Unlike related GEFs, DENND1C uses a unique C-terminal motif to directly bind actin, thereby coupling Rab35 activation to the actin cytoskeleton and resulting in changes to cell shape such as the formation of membrane protrusions[3]. While all three DENND1 family members (DENND1A–C) are GEFs for Rab35, DENND1C is characterized by a preference for actin-related processes, and has also shown low activity towards Rab8A and Rab13[3][4]. The gene is implicated in cellular proliferation, apoptosis, and receptor signaling, and associated diseases include congenital disorders of glycosylation type Iic[4]. Its knockout models are used in the exploration of gene function, particularly for understanding its role in health and disease, though no direct pharmacological agents targeting DENND1C or established biomarker or safety data are currently documented[1][4].
