Overview

DENN domain-containing protein 4B (DENND4B) is a cytoplasmic and membrane-associated guanine nucleotide exchange factor (GEF) that functions directly in the regulation of Rab protein signal transduction, especially Rab10. It promotes the exchange of GDP for GTP, switching Rab proteins from their inactive to active forms and supporting vesicle trafficking through the endomembrane system, with main localization in the Golgi apparatus and nucleoplasm. Human DENND4B has been linked with rare inherited disorders and is expressed in many tissues. Although some cancer-related data exist, DENND4B is not yet an established therapeutic target, and no drugs specifically target this protein[1][4][10][6][7].
