Overview

DENN domain-containing protein 5B (DENND5B) is a large (~145 kDa) guanine nucleotide exchange factor (GEF) that activates Rab GTPases, orchestrating GDP-GTP exchange to regulate vesicular trafficking, particularly in chylomicron secretion from the Golgi to the plasma membrane. It contains canonical DENN domains, two RUN domains (involved with cytoskeletal dynamics and Rab regulation), and a PLAT domain (associated with membrane interactions). DENND5B is highly expressed in the brain, small intestine, and liver, and is essential for lipid absorption and neural development. Functionally, it interacts with MYO5A (myosin-Va) and valosin-containing protein (VCP), supporting actin-based secretory vesicle transport and protein quality control. Genetic variation in DENND5B influences metabolic traits such as plasma lipids and body mass index, and aberrant expression is linked to cancer and neurodevelopmental disorders. It is increasingly recognized as a potential prognostic biomarker and an emerging molecular target in metabolic and oncological contexts.
