Overview

Density separation media are specialized, often inert chemical reagents used in laboratory and clinical centrifugation protocols to isolate specific biological components—such as cells, organelles, or viruses—from complex mixtures based on their buoyant density. These media, which include substances such as Ficoll (a high molecular weight sucrose polymer), Percoll (silica particles coated with polyvinylpyrrolidone), and iodixanol, facilitate the formation of density layers that allow for the stratification of components during centrifugation (Sigma-Aldrich, 2023). For instance, Ficoll-Paque is widely utilized as the gold standard for isolating peripheral blood mononuclear cells (PBMCs) from whole blood, separating lymphocytes and monocytes from denser erythrocytes and granulocytes (Cytiva, 2022). It is critical to recognize that density separation media are not therapeutic targets; rather, they are physical tools essential for diagnostic processing, vaccine manufacturing, and basic research. While designed for biocompatibility, these reagents can exert osmotic pressure or physical stress on delicate samples, potentially influencing the physiological state of the isolated material and requiring validation for downstream applications (Boyum, 1968).
