Overview

Dentin is a calcified connective tissue that constitutes the majority of the tooth structure, located beneath the enamel in the crown and cementum in the root; it surrounds the pulp chamber and provides the tooth’s basic shape and rigidity[3][5][7]. Dentin consists mostly of hydroxyapatite crystals (about 70% of its dry weight), a collagenous organic matrix, and water[3][5]. One defining feature of dentin is the presence of dentinal tubules—microscopic channels radiating from the pulp to the external boundaries of the tooth. These tubules contain cellular projections from odontoblasts, dentinal fluid, and facilitate nerve signal conduction, thereby playing a role in tooth sensitivity and pain transmission when exposed due to enamel or cementum loss[1][5][7]. Dentin is produced by odontoblasts and can regenerate to some extent via the formation of secondary or tertiary dentin in response to injury. It is less mineralized than enamel but tougher and harder than bone. Dentin has no pharmaceutical targets but is important in dental medicine due to its role in tooth structure and sensitivity. While various non-collagenous proteins such as dentin sialophosphoprotein (DSPP), dentin phosphoprotein (DPP), and dentin glycoprotein (DGP) are found in the organic matrix and are critical for mineralization and development, the tissue itself cannot be considered a “target molecule/receptor” in the conventional therapeutic sense[2][4][6][9]. It is important to note that Dentin is a tissue, not an individual molecular drug target or receptor. Proteins like DSPP, DPP, and DGP, found in dentin, are sometimes studied extensively but are not themselves traditional therapeutic targets at this time. “Dentin” may be confused with dentin matrix proteins in molecular contexts, but as queried, it does not fit the criteria for inclusion in databases of drug targets, receptors, or enzymes. While dental materials and medicaments may interact with or affect dentin, these are not considered drug-receptor interactions in particularly in the context of typical biomolecule targeting.
