Overview

Dentinal tubule openings are microscopic apertures at the ends of dentinal tubules, which run through the dentin from the pulp chamber toward the enamel or cementum. These openings serve as conduits for fluid, nutrients, and cellular processes—specifically, the extensions of odontoblasts, sensory nerves, and extracellular matrix—which together mediate mechanical and sensory functions within the tooth. Fluid movement through tubule openings enables mechanotransduction, allowing odontoblasts to respond to external stimuli, and importantly, forms the physiological basis for tooth sensitivity and pain. Exposure of these openings through enamel erosion or dental disease increases the risk of pulp irritation, sensitivity, and ingress of pathogens, making them central to oral health, though not a classic drug target in molecular pharmacology.
