Overview

The dentin tubule is a microscopic channel in the dentin layer of teeth, running from the pulp to the outer surface. It is lined by a complex organic matrix, mostly collagen and proteoglycans, housing odontoblast processes and transmitting sensory stimuli (pain, pressure). The tooth enamel mineral matrix is the outermost, hardest layer of the tooth, composed of 95–96% mineral (hydroxyapatite) and a small fraction of organic matrix proteins, mainly during tooth development. Enamel matrix proteins such as amelogenin, enamelin, and ameloblastin form an early scaffold for mineral deposition, but mature enamel is mostly mineral with minimal organic matter[1][2][3][4][6][7][8][9][10]. Together, these matrices support tooth structure and function, but neither is a molecular target for drugs in the classic sense. Their integrity is essential for resistance to caries and sensitivity. Interventions typically aim to preserve or restore their mineral content and structure through topical agents or remineralization therapies.
