The dentin tubule surface is the internal lining of the microscopic channels that traverse the dentin layer of the tooth, connecting the dental pulp to the enamel or cementum (Pashley, 1996). These tubules contain odontoblastic processes and dentinal fluid, serving as the primary pathway for sensory transduction and nutrient transport within the tooth (Brännström, 1963). In clinical conditions such as dentin hypersensitivity, the exposure of the dentin tubule surface allows external stimuli to cause rapid fluid movement, which activates pulpal nociceptors according to the hydrodynamic theory (Brännström, 1963; Liu et al., 2020). Therapeutic interventions target this surface by either physically occluding the tubule orifices with minerals like calcium sodium phosphosilicate or stannous fluoride, or by using chemical agents like potassium nitrate to desensitize the underlying nerves (Miglani et al., 2010). While the surface itself is an anatomical structure rather than a single protein or receptor, it is the functional site for various desensitizing and remineralizing pharmacological agents (Miglani et al., 2010; Pashley, 1996).
Therapeutic agents act by either physically occluding the tubule orifices to prevent hydrodynamic fluid movement or by chemically depolarizing intradental nerve fibers to inhibit pain signal transmission.
