Overview

Dentinal collagen fiber refers to the network of primarily type I collagen fibrils that form the organic scaffold of dentin, the mineralized tissue beneath tooth enamel. These fibers constitute the bulk of the organic content in dentin, comprising about 90% type I collagen with smaller amounts of types III and V, and provide mechanical strength and resilience by serving as the structural framework for mineral deposition with hydroxyapatite during dentinogenesis[1][2][3][4]. Collagen fibers are oriented in bundles and help organize the hierarchical architecture of dentin, forming intertubular and peritubular regions central to dentin’s mechanical function and response to injury[2][3][4]. They do not possess direct receptor, enzyme, or transporter activity. Degradation of these fibers, mainly by matrix metalloproteinases (MMPs) and other endogenous enzymes, undermines the mechanical integrity of dentin, and is a fundamental process in conditions like caries and in the failure of dental restorations[1][3]. They are not considered classical therapeutic targets but are critical in dental biomaterials research, tissue engineering, and the development of adhesives that interact or protect the collagen during restorative procedures[1][2][3][4].
