Overview

Dental enamel and dentinal tubules are the primary structural components of the human tooth crown. Enamel is the highly mineralized, acellular outer layer composed mainly of hydroxyapatite crystals, providing a hard protective barrier against mechanical and chemical wear (StatPearls, 2023). Dentinal tubules are microscopic channels within the underlying dentin layer that house odontoblastic processes and fluid. They play a critical role in tooth sensitivity through the hydrodynamic theory, where fluid movement triggers nerve fibers in the pulp (PubMed, 2021). While not traditional molecular targets like receptors or enzymes, these structures are the focus of dental therapies aimed at remineralization and desensitization. Drugs such as fluoride interact with the enamel surface to form acid-resistant fluorapatite (NIH, 2022). Desensitizing agents like potassium nitrate or tubule-occluding compounds (e.g., arginine, calcium sodium phosphosilicate) target the tubules to alleviate pain (PubMed, 2020). This target is considered "incorrect" in a pharmacological context because it represents a complex anatomical tissue rather than a specific protein or molecular entity.
