Overview

Deoxycytidine kinase (dCK)-independent cytotoxicity via gemcitabine monophosphate release refers to a strategy to induce cell death using gemcitabine without relying on the canonical activation step mediated by dCK. This involves delivering gemcitabine monophosphate (dFdCMP) directly or through prodrugs, bypassing dCK and overcoming resistance in tumors with low or absent dCK activity. The downstream effects include incorporation of dFdCTP into DNA, leading to masked chain termination, and inhibition of ribonucleotide reductase by dFdCDP.
