Overview

Deoxyguanosine kinase antisense RNA 1 (DGUOK-AS1) is a long non-coding RNA (lncRNA) located on chromosome 2q24.32, composed of two exons with a total transcript length of 563 nucleotides[1][3]. DGUOK-AS1 shows negligible protein-coding potential and functions as an antisense RNA to the DGUOK gene. Recently, DGUOK-AS1 has been identified as an oncogenic lncRNA in several human cancers, including breast cancer, non-small cell lung cancer, and cervical cancer[1][4]. It is usually upregulated in tumor tissues, and its high expression correlates with poorer prognosis and increased tumor aggressiveness in breast cancer patients[1][3]. Functionally, DGUOK-AS1 can act as a competing endogenous RNA (ceRNA), sponging tumor-suppressive microRNAs (such as miR-497-5p) to derepress oncogenic mRNAs, thereby modulating gene networks involved in cell proliferation, migration, invasion, and angiogenesis[1][3][4]. In NSCLC, DGUOK-AS1 promotes tumor cell proliferation and metastasis by stabilizing TRPM7 mRNA via the METTL3/IGF2BP2 pathway in an m6A-dependent manner[4]. DGUOK-AS1 is primarily cytoplasmic but may interact with regulatory proteins and RNAs, playing an integral role in tumor growth and spread. Its prominent expression and functional significance make it a candidate prognostic biomarker and a potential therapeutic target, although no approved drugs are currently known to directly target DGUOK-AS1[1][3][4].
