Deoxyhypusine synthase (DHPS)

Target
DHPS
Molecular classification
Enzyme, Transferase
01

Overview

Deoxyhypusine synthase (DHPS) is an essential enzyme that catalyzes the first step in the post-translational modification of eukaryotic initiation factor 5A (eIF5A), specifically forming deoxyhypusine, a precursor to hypusine—a unique amino acid found only in eIF5A. This modification is critical for the activation and function of eIF5A, which plays a central role in mRNA translation and cellular growth. DHPS is considered a therapeutic target in cancer, diabetes, and infectious diseases.

02

Mechanism of action

Inhibition of DHPS leads to reduced hypusination of eIF5A, thereby impairing mRNA translation and cellular growth/division.

Safety considerations

Inhibition of DHPS may affect normal cellular processes due to its essential role in protein synthesis.Off-target effects need to be considered when developing DHPS inhibitors.
