Overview

Deoxynucleotidyltransferase terminal-interacting protein 1 (DNTTIP1) is a multifunctional, chromatin-associated protein that enhances the DNA polymerase activity of terminal deoxynucleotidyltransferase (DNTT) and acts as a transcriptional regulator by binding DNA and nucleosomes and recruiting histone deacetylase (HDAC) complexes to chromatin[1][2][4][5]. DNTTIP1 contains AT-hook and helix–turn–helix motifs, enabling it to bind AT-rich genomic regions and regulate the expression of key genes such as RAB20[2]. It plays roles in transcriptional silencing, cell cycle control, and developmental signaling, and is implicated in the modulation of tumor suppressor p53 via deacetylation. Overexpression or dysregulation of DNTTIP1 is associated with increased cell proliferation and poor prognosis in several cancers (e.g., hepatocellular carcinoma, oral squamous cell carcinoma, non-small cell lung cancer). Additionally, it can alter immune cell infiltration and function in the tumor microenvironment[4]. The protein has a dimerization domain and interacts with other chromatin modifiers, making it an emerging target of interest for biomarker studies, though currently no drugs or direct modulators are reported[1][2][4][5].
