It catalyzes the hydrolysis of DNA by cleaving phosphodiester bonds, contributing to the degradation of nuclear DNA primarily during keratinocyte differentiation in the skin[1]. This activity is crucial for the removal of nuclear material during cornification and the formation of the skin barrier[1]. DNASE1L2 is conserved in amniotes and, unlike other Dnase1 family members that are secreted, its principal action is intracellular[1][5]. The enzyme is predicted to be activated following organelle disruption, allowing it access to nuclear DNA, although detailed mechanisms of escape from the endoplasmic reticulum or lysosome remain unclear[1]. DNASE1L2 does not appear to be a common direct therapeutic target and there are no reported drugs or biomarkers associated specifically with this enzyme[4][5]. Its dysfunction or altered expression may contribute to skin disorders associated with abnormal DNA degradation[1].
