Overview

Deoxyribonuclease-2-beta (DNASE2B) is a lysosomal endonuclease enzyme that catalyzes the hydrolysis of double-stranded DNA at acidic pH and does not require divalent cations for activity[1][2][6]. It participates in the degradation of nuclear DNA, notably during the differentiation of lens cells in the eye and is mainly expressed in the salivary gland and lungs in humans[1][2][6]. DNASE2B is structurally related to DNASE2, but has a much more restricted tissue expression[1][2][6]. Genetic variants in DNASE2B have been associated with ocular disease, particularly cataract[1]. DNASE2B is part of the DNase2 family of lysosomal enzymes involved in the removal of self-DNA from dying cells and the maintenance of tissue homeostasis[4]. No drugs are currently known to target DNASE2B directly, and its primary characterized function is in physiological DNA clearance rather than pathology or direct therapeutic intervention.
