Deoxythymidylate kinase (DTYMK) is a pyrimidine metabolic enzyme that catalyzes the phosphorylation of deoxythymidine monophosphate (dTMP) to deoxythymidine diphosphate (dTDP), a key step in the synthesis of deoxythymidine triphosphate (dTTP), which is essential for DNA replication and repair. DTYMK is vital for cell cycle progression and genome stability; its deficiency causes severe neurodevelopmental diseases, while its overexpression is linked to cancer progression due to increased nucleotide synthesis capacity for rapidly dividing cells[1][2][3].
Inhibition leads to depletion of dTTP, DNA synthesis impairment, and cell death, particularly in highly proliferative cells such as cancer cells[1][2]
