Overview

DEP domain containing 1 (DEPDC1) is a cancer-testis antigen that is significantly overexpressed in a variety of human malignancies, including bladder cancer and non-small cell lung cancer, while remaining nearly absent in normal adult tissues except for the testis (Obara et al., 2012, Cancer Science). The DEPDC1-derived peptide presented by the Human leukocyte antigen A*24:02 (HLA-A*24:02) allele serves as a specific target for cancer immunotherapy, particularly in populations where the HLA-A*24:02 genotype is prevalent (Yuan et al., 2014, Molecular Medicine Reports). This peptide-MHC complex is recognized by the T-cell receptors (TCRs) of cytotoxic T lymphocytes (CTLs), which can then selectively eliminate cancer cells expressing the antigen. Therapeutic approaches targeting this complex primarily include peptide-based vaccines, such as the DEPDC1-9-294 peptide, designed to elicit a robust, antigen-specific immune response (NCT00910598). Clinical studies have demonstrated that vaccination with DEPDC1-derived peptides can induce specific CTL responses and may correlate with improved clinical outcomes in patients with advanced cancers. Because of its high tumor specificity and restricted expression in normal tissues, the DEPDC1-HLA-A24:02 complex represents a promising target for precision oncology and personalized immunotherapy.
