DEP domain-containing protein 1 (DEPDC1) is a cancer-testis antigen that is significantly upregulated in various malignancies, particularly bladder cancer and non-small cell lung cancer, while showing minimal expression in normal adult tissues except the testis (Obara et al., 2012; PubMed: 22433374). The DEPDC1-9-294 peptide (sequence: EYYELGVHV) is a 9-mer epitope derived from the DEPDC1 protein that is specifically presented by the HLA-A*24:02 allele (Yuan et al., 2014; PubMed: 24583115). This peptide-MHC complex serves as a target for cancer immunotherapy, including peptide-based vaccines and T-cell receptor (TCR) engineered therapies. Clinical studies have demonstrated that vaccination with this peptide can induce specific cytotoxic T lymphocytes (CTLs) that recognize and kill DEPDC1-expressing tumor cells in HLA-A24-positive patients (Taniura et al., 2012; PubMed: 22314581). The restricted expression pattern of DEPDC1 makes this epitope a promising target for minimizing off-target toxicity in oncology treatments. Ongoing research continues to evaluate its efficacy in combination with other immunomodulators to enhance the anti-tumor immune response.
