Dermatan sulfate (DS) and heparan sulfate (HS) are complex, sulfated linear polysaccharides belonging to the glycosaminoglycan (GAG) family, found ubiquitously on cell surfaces and in the extracellular matrix (UniProt). They function as critical co-receptors and modulators for various proteins, including growth factors, cytokines, and protease inhibitors like antithrombin III, thereby regulating cell signaling, tissue morphogenesis, and blood coagulation (PubMed: 25631224). Pathologically, the impaired degradation of DS and HS leads to their accumulation in lysosomes, resulting in Mucopolysaccharidosis (MPS) types I, II, and VI, which manifest as multi-systemic organ damage (NIH: GARD). These molecules are therapeutic targets in several contexts: as anticoagulants (e.g., heparin and danaparoid) that enhance the inhibition of clotting factors, and as substrates for enzyme replacement therapies (e.g., laronidase, idursulfase) designed to reduce toxic accumulation in MPS patients (StatPearls: NBK538247). Additionally, HS and DS are being investigated as targets for blocking viral entry and inhibiting tumor metastasis due to their roles in pathogen binding and cancer cell signaling (PubMed: 32533971).
