Dermatophagoides farinae allergens are a group of potent proteins derived from the American house dust mite, primarily found in mite fecal pellets and decaying body parts. These allergens, most notably the major allergens Der f 1 (a cysteine protease) and Der f 2 (a lipid-binding protein), are among the most common triggers for IgE-mediated hypersensitivity reactions globally. In sensitive individuals, inhalation of these proteins activates mast cells and Th2-driven immune pathways, leading to chronic inflammatory conditions such as allergic rhinitis, asthma, and atopic dermatitis. Therapeutically, these allergens serve as the active components in allergen-specific immunotherapy (AIT), such as the FDA-approved sublingual tablet ODACTRA. By exposing the patient to gradually increasing or sustained low doses of the allergen, these treatments aim to desensitize the immune system, inducing long-term tolerance and reducing the clinical severity of allergic symptoms upon natural exposure.
Allergen-specific immunotherapy (AIT) involves the repeated administration of specific allergens to patients with IgE-mediated conditions to induce immunological tolerance. This process shifts the immune response from a Th2-biased allergic profile toward a Th1/Treg profile, leading to the production of blocking antibodies such as allergen-specific IgG4 and the suppression of mast cell and basophil activation.
